ABSTRACT

This study used metabolomics to explore the improvement effect of raw and honey-processed Glycyrrhizae Radix et Rhizoma on acute kidney injury (AKI) in rats. All animal experiments were approved by the Animal Ethics Committee of Shandong Academy of Chinese Medicine (approval No.: SDZYY20200101001). SD rats were randomly divided into normal group, model group, raw Glycyrrhizae Radix et Rhizoma group (0.9 g·kg-1) and honey-processed Glycyrrhizae Radix et Rhizoma group (0.9 g·kg-1), 6 rats in each group. The rats model of acute kidney injury was established by single intraperitoneal injection of cisplatin (CP) and treated with raw and honey-processed Glycyrrhizae Radix et Rhizoma. The pathological changes of renal tissue were evaluated by hematoxylin and eosin (HE) and PAS staining, the contents creatinine (Cr), blood urea nitrogen (BUN) and superoxide dismutase (SOD) in serum were detected. UPLC-Q-TOF/MS was used to study tissue metabolomics to screen the biomarkers affected by raw and honey-processed Glycyrrhizae Radix et Rhizoma and analyz the metabolic pathways. The results showed that compared with the model group, raw and honey-processed Glycyrrhizae Radix et Rhizoma can significantly improve the pathological changes of renal tissue and decrease the content of Cr, BUN and increase the activity of SOD. In addition, honey-processed Glycyrrhizae Radix et Rhizoma can also significantly reduce the kidney index. In tissue samples, 45 biomarkers were measured in AKI rats. Raw Glycyrrhizae Radix et Rhizoma and honey-processed Glycyrrhizae Radix et Rhizoma could simultaneously call back 11 differential metabolites, which were involved in the regulation of glycerophospholipid metabolism, tryptophan metabolism, phenylalanine, tyrosine and tryptophan biosynthesis, and glutathione metabolism. In addition, raw Glycyrrhizae Radix et Rhizoma is also involved in the regulation of glycine, serine and threonine metabolism and pyrimidine metabolism. In summary, raw and honey-processed Glycyrrhizae Radix et Rhizoma can participate in the regulation of different metabolic pathways, and play an improvement role in AKI rats by regulating amino acid, lipid metabolism, energy metabolism and oxidative stress.

ABSTRACT

Objective To investigate the application value of eye tracking in lie detection. Methods The 40 subjects were randomly divided into two groups. The pupil diameter, fixation duration, points of fixation and blink frequency of the subjects in the experimental group in observing target stimulation and non-target stimulation were recorded with eye tracker after they accomplished the mock crime. The eye movement parameters of subjects in the control group were directly collected. The differences in eye movement parameters of the experimental group and the control group in observing target stimulation and non-target stimulation were analyzed by t-test. Pearson coefficient analysis of correlation between eye movement parameters that had differences was conducted. The effectiveness of eye movement parameters to distinguish between the experimental group and the control group was calculated by the receiver operator characteristic (ROC) curve. Results Participants from the experimental group had shorter average pupil diameter, longer average fixation duration and fewer fixation points (P<0.05), but the differences in blink frequency had no statistical significance. The differences in the above indicators of the control group in observing target stimulation and non-target stimulation had no statistical significance. The average fixation duration showed a negative correlation with fixation points (r=-0.255, P<0.05); the average fixation duration showed a negative correlation with average pupil diameter (r=-0.218, P<0.05); the fixation points showed a positive correlation with average pupil diameter (r=0.09, P<0.05). The area under the curve of average pupil diameter, average fixation duration and fixation points was 0.603, 0.621 and 0.580, respectively. Conclusion The average pupil diameter, average fixation duration and fixation points obtained by the eye tracker under laboratory conditions can be used to detect lies.

ABSTRACT

BACKGROUND: In recent years, because of its wide range of sources, easy access, strong self-renewal capacity, multi-directional differentiation potential, low immunogenicity and low ethical controversy and other characteristics, human Wharton's Jelly-derived mesenchymal stem cells have become popular seed cells in regenerative medicine. OBJECTIVE: To review the advances in directional differentiation of human Wharton's Jelly-derived mesenchymal stem cells in recent five years. METHODS: The PubMed and CNKI databases were searched by the first author using the keywords of "Wharton Jelly derived mesenchymal stem cell, human umbilical cord mesenchymal stem cell, differentiation" in English and Chinese, respectively. The retrieval time was from January 2012 to January 2017. After initial search, 120 articles were collected, and 46 articles were included in final analysis. RESULTS AND CONCLUSION: Up to now, there are more than 18 types of cells that have been differentiated from Wharton's Jelly-derived mesenchymal stem cells. In recent 5 years, some studies have improved the previous induction method, some studies have developed the new directional differentiation of Wharton's Jelly-derived mesenchymal stem cells, and the others have explored the involved signal pathways and relevant molecular mechanisms. Differentiated Wharton's Jelly-derived mesenchymal stem cells can be used for human tissue regeneration and repair, which give hope to the treatment of many refractory diseases.

ABSTRACT

Brown algae are well known as a source of biologically active compounds, especially those having antioxidant activities, such as phlorotannins. In this study we examined the antioxidant activities of crude phlorotannins extracts (CPEs) obtained from Sargassum hemiphyllum (SH) and fractionated according to the molecular weights. When CPEs were administrated at a dose of 30 mg/kg to Kunming mice pre-treated with carbon tetrachloride (CCl4), the levels of oxidative stress indicators in the liver, kidney and brain were significantly reduced in vivo. All the components of various molecular weight fractions of CPEs exhibited greater scavenging capacities in clearing hydroxyl free radical and superoxide anion than the positive controls gallic acid, vitamin C and vitamin E. Particularly, the components greater than 30 kD obtained from ethyl acetate phase showed the highest antioxidant capacities. These results indicated that SH is a potential source for extracting phlorotannins, the algal antioxidant compounds.

ABSTRACT

<p><b>OBJECTIVE</b>To explore the effects of high dose glycosides of Tripterygium wilfordii Hook. f (GTW) on the fertility of young rats.</p><p><b>METHODS</b>Fifty female SD young rats and 50 male SD young rats were randomly divided into the blank group and the GTW group, 25 in each. GTW was given at the daily dose of 9 mg/kg. After 12 weeks of medication, the male rats were caged together with healthy adult female rats in the ratio of 1:1. The female rats were caged together with healthy adult male rats in the ratio of 2:1. The cage process lasted for two weeks, totally for three times. The pregnant rate of female rats and the survival rate of baby rats were then observed.</p><p><b>RESULTS</b>There was no significant difference in the pregnant rate or the survival rate of baby rats in the GTW group.</p><p><b>CONCLUSION</b>High dose GTW showed no obvious effects on the fertility of adult rats or the growth and development of new born rats.</p>

ABSTRACT

Objective To explore the pathologic feature of children with kidney disease.Methods A retrospective analysis on renal bio-psy findings in 246 cases of children patients.Pathological classification was made according to the modified WHO criteria of 1995 for renal pathology.Results Of the 246 children,104 cases were diagnosed as primary glomerulonephritis,accounting for 42.28% of the total cases,136 cases as secondary glomerulonephritis,accounting for 42.28%,3 cases as hereditary nephritis,accounting for 1.22%,and 3 cases as unclassified renal disease,accounting for 1.22%.In primary glomerulonephritis,66 cases were diagnosed as nephrotic syndrome,23 cases as persistent glomeruloneplritis,8 cases as acute nephritis syndrome,3 cases as chronic nephritis syndrome,2 cases as isolated proteinuria,1 case as rapidly progressive glomerulonephritis,1 case as isolated hematuria.IgA nephropathy was the most frequent pathological type,accounting for 15.85%(39 cases),followed by mesangial proliferative glomerulonephritis,minimal change disease,endocapillary proliferative glomerulinephritis,IgM nephropathy,membranous nephropathy,focal segmental glomerulosclerosis,and minor lesion nephropathy.In secondary glomerulonephritis,Henoch-Schonlein purpura nephritis accounting for 48.37%(119 cases),followed by hepatitis B virus associated nephritis(11 cases) and lupus nephritis(6 cases).In hereditary nephritis,there were 2 cases with thin glomerular basement disease and 1 case with Alport syndrome.Conclusions Among the 246 cases of renal biopsy data,the secondary glomerulonephritis,especially Henoch-Schonlein purpura nephritis,is more common than primary glomerulonephritis.In primary glomerular diseases,IgA nephropathy is the most frequent pathological type.
